Note: In
PMID:17855365
, to determine native phosphorylation sites, FSCB was immunoprecipitated from mouse sperm, separated by SDS-PAGE. A 270-kDa band immunoreactive with anti-FSCB serum was cored from a parallel silver stained gel and subjected to mass spectrometric phosphopeptide analysis. This produced evidence of 7 different sites, however for one of them, there are two possible sites within the same singly phosphorylated phosphopeptide, so the analysis does not allow determination between Ser-161 and Thr-163. Two of these sites observed to be phosphorylated in vivo are canonical PKA sites (Rx[ST]). In vitro analysis of recombinant FSCB indicates that it is a substrate for purified PKA, but does not indicate which sites.
